Apta-sensors for spore detection
An aptamer-based biosensor was developed to detect B. cereus spores in milk. DNA aptamers are small oligonucleotides characterized by high stability and morphological specificity for the targets and may bind whole bacterial cells and spores. Spores of pathogenic microorganisms represent a main concern for food safety, due to their high resistance to treatments. The classical methods for spore detection are laborious, time consuming and food matrices as milk represent additional difficulties due to their complexity. Aptamer-based biosensors offer a rapid, highly specific and low cost detection directly in food matrices. The specificity of the aptamers was firstly confirmed and the sensor construction optimized. Detection of spores was tested both in a buffer and milk. Promising results were obtained, with a theoretical LOD of 3 cfu/ml in the food matrix. To detect spores of different strains of B.cereus group, we applied a mix of three aptamers targeting different epitopes on the spore surface. However, using this strategy the sensibility of the detection decreased compared to that obtained using a single aptamer.
